Description:
Sequence tagged Ds insertion line, generated by Agrobacterium tumefaciens root transformation of Landsberg erecta plants; kanamycin was employed for selection of plants carrying a Ds insertion (50ug/ml); potential gametophytic mutants were selected based on kanamycin segregation ratios <2:1. Reciprocal crosses were used to estimate female and male gametophytic transmission. Genomic DNA flanking the transposon insertion in each line was amplified by PCR and directly sequenced. The Ds insertion line was crossed as female with the qrt1 homozygous mutant in the Ler background. Resistant F2 plants that carry the Ds insertion were screened for the qrt1 phenotype. Backcrossed 2 times to Ler-0. Insertion: 141 bp upstream
