Quadruple mutant: the ahp1-1 (CS860143) mutant isolated in the Ws ecotype was crossed to the Col ecotype twice before being crossed to the single mutant ahp2-1 (CS860144) isolated in the Col ecotype, generating the double mutant ahp1-1,2-1(CS860150). The ahp3 (CS860145) mutant was crossed to ahp4 (CS860146) to generate the double mutant ahp3,4 (CS860154). These two double mutants were crossed to generate the quadruple mutant ahp1-1,2-1,3,4.

Inhibition of root elongation is substantially less sensitive to cytokinin than the wild type plants and is less responsive to cytokinin over the whole range of cytokinin benzyl adenine (BA) concentrations used in the growth media; roots are longer than those of wild-type plants when grown on near-saturating levels of cytokinin (10 uM BA). It is not significantly different from the response of the ahp1,2,3 triple mutant, which indicates that AHP4 does not play a substantial role in this response; reduced inhibition of hypocotyl elongation in response to cytokinin, consistent with reduced sensitivity to the hormone.
