Description:
CRISPR-Cas9-mediated mutation. A gRNA that directs the NAC050 and NAC052 common target sequence: GAAGGTTAAT-GATCTCCAGA was produced, and the psgRNA-CAS9-At fragment was inserted into pCAMBIA1300. In the nac050/052 double mutant, a cytosine nucleotide is separately inserted into both NAC050 or NAC052 and leads to frame shifts in both genes (Supp. Figure S10 of PMID: 25578968). Original donated stock: Homozygous double mutant seeds of T4 lines, with the transgene segregated out.
