Description:
Consists of a 9.1 kb insert into the broad host range cosmid pRK290 (Ditta et al., 1980). The insert carries the cos site (for lambda packaging), a 35S-NPTII gene (confering kanamycin resistance), and a dark Bluescript (dBS) polylinker between the T-DNA borders. The dBS polylinker provides a number of unique restriction enzyme sites for cloning and gives a darker than usual blue colour to E. coli colonies on X-Gal and IPTG, which is an advantage in a low copy binary vector. pRK290 carries a bacterial tetracycline resistance gene.
Growth Requirement: grow in LB media at 37 C overnight
