Description:
Nitrate reductase (NR)-null mutant; double mutant constructed from two lines that had null mutations in either the NIA1 or the NIA2 NR gene; the first line (nia2-5, also named chl3-5, Columbia background) had a deletion in the NIA2 gene, the second line (nia1::Ds or nia1-2, Ler background) had a Ds insertion in the NIA1 gene; no detectable NR activity and can not use nitrate as the sole nitrogen source; requires ammonia as nitrogen source.
Growth Requirement: germinate on agarose plates (2.5 mM ammonium succinate, pH 6.5), transplant to autoclaved vermiculite pots, irrigate with medium (pH6.5) with 2.5 mM ammonium succinate as sole nitrogen source.
