Description:
ADH1 entry vectors containing left and right zinc finger nucleases (ZFNs) were recombined with pMDC7, giving rise to the T-DNA expression vector pFZ62, which was transformed into A.tumefaciens strain GV3101. Transformed Agrobacterium was used for floral dip transformation of Col plants. Transformed seeds were grown on Hygromycin as a selection marker and 17 beta-estradiol to induce the expression of the ZFN. T1 lines for which the mutation in the ADH1 gene was confirmed by sequencing were grown to maturity and T2 seeds were collected. Donated seeds represent T3 generation. May segregate ZFN transgene.
