Description:
To create a transgenic line in which nearly all nuclei can be captured with the INTACT method, we transformed A. thaliana accession Col-0 with a construct (UBQ10:NTF) in which the UBQ10 promoter was fused with the NTF segment used in the original INTACT lines. We simultaneously transformed the UBQ10:NTF construct and the ACT2:BirA construct by floral dip to obtain a stable homozygous UBQ10:NTF::ACT2:BirA line. The UBQ10:NTF construct confers Kanamycin resistance at 50 ug/mL for plated seedlings; the ACT2:BirA construct confers BASTA resistance at 15 uM for plated seedlings.
