Description:
Isolated from a population of EMS-mutagenized M2 Ler-0 seeds from Lehle Seeds in a screen for mutants with altered giant cells in the sepal. Heterozygous for the mutation. Backcrossed 3 times to Ler-0. The sec24a-2 mutation can be genotyped by amplifying with primers: 5 -CAT AGC TCC AAC TGA ATA CAT GGT TCG GAC T-3 and 5 -AAG TGG GAT ATT CAA ATC GAT CAT GCT-3 at an annealing temperature of 55C, followed by digesting the product with HinfI to produce a 102-bp wild-type product or a 130-bp mutant product.
