Description:
Transgenic line; 1,511-bp encompassing the PDH1 promoter fragment out to -1,389 bp and the 5 UTR (122 bp) of the PDH1 transcript was amplified and ligated into the pJET1.2 cloning vector, then subcloned into pCAB2-LUC2-pCAMBIA1390 containing the pGL4.10-LUC2 Luciferase2 sequence. It was further digested with SalI/PstI to remove the CAB2 promoter and was then ligated with the PstI-PDH1promoter-SalI fragment. The Col-0 plant was transformed using the floral dip method. Hygromycin resistant. Homozygous for the transgene.
