Description:
Transgenic GFP-fusion expression line harboring the pS27(AT1G68130)::cS27:GFP(-G):NOS #1 construct. The GFP reporter fusion construct was generated by using the MultiSite Gateway Three-Fragment Vector Construction system (Invitrogen); final construct in Agrobacterium was transformed into Columbia ecotype plants by using the floral dip method; transformed plants (T1) were selected with Basta on soil and 5 to 7-day-old T2 seedlings were grown and imaged by using confocal laser scanning microscopy; distributed seeds are T3.
Growth Requirement: Segregating for the transgene. basta resistant.

Transcription factor (TF) expressed in a tissue-enriched manner in roots: nuclear localized GFP expression in the pericycle starting from late meristimatic zone (only in the pericycle of xylem pole up to early maturation zone) very weak GFP in endodermis and cortex from elongation zone(4/5), also very strong GFP in endodermis and cortex, possibly protein is moving (2/5; 5DAG).
