Description:
Transgenic GFP-fusion expression line harboring the pS8(AT5G60200)::cS8:GFP(-G):NOS construct. The GFP reporter fusion construct was generated by using the MultiSite Gateway Three-Fragment Vector Construction system (Invitrogen); final construct in Agrobacterium was transformed into Columbia ecotype plants by using the floral dip method; transformed plants (T1) were selected with Basta on soil and 5 to 7-day-old T2 seedlings were grown and imaged by using confocal laser scanning microscopy; distributed seeds are T3.
Growth Requirement: Segregating for the transgene. basta resistant.

Transcription factor (TF) expressed in a tissue-enriched manner in roots: nuclear GFP expression in the pericycle of the phloem pole starting from above the quiescent center to the hypocotyl (2/6); nuclear GFP in the stele of the promeristem, no GFP in the late meristematic and early elongation zone but GFP appears from late elongation zone which is cytoplasmic and spotty (2/6; 5DAG).
