Description:
fer-4 mutant complemented with kinase-dead FER. The genomic FER sequence containing the native promoter, coding sequence, and 3 -UTR was amplified and cloned into the pCR8/GW/TOPO vector. Site-directed mutagenesis was performed to introduce a mutation at Lys 565 to Arg. The LR clonase reaction was used to transfer the FER-K565R clone into the pMDC99 plant transformation vector. Heterozygous fer-4 plants were transformed and transgenic plants were selected on hygromycin. Plants homozygous for the fer-4 allele and single insertion of the transgene were isolated by genotyping.
