Description:
pif3-3 mutant expressing 35S::6xHis:PIF3:5xMYC. PIF3 cDNA was amplified with a forward primer containing 6xHis and a reverse primer containing 1xMYC. The product was cloned into the pENTR/D-TOPO vector and transferred into the pGWB17 binary vector containing a 35S promoter and 4xMYC fusion at the C terminus. The construct was then transformed into pif3-3 mutants plants and transgenic plants were screened in GM medium containing hygromycin and kanamycin. Homozygous for the transgene.
