Description:
Transgenic line expressing self-resistance gene astD. A 35-amino acid residue chloroplast localization signal (CLS) derived from the endogenous A. thaliana DHAD gene was fused to the N-terminus of an A. terreus astD gene codon-optimized for A. thaliana, and a 3xFLAG-tag was inserted in between. The construct was cloned into a pEG202 vector modified to express the transgene from the UBQ10 promoter. Col-0 plants were transformed by the floral dip method. BASTA and aspterric acid resistant. Segregating for the transgene. Independent transgenic line 2; original donated stock is from the T2 generation.
Growth Requirement: Positive transgenic plants can be selected with glufosinate and testing for survival in the presence of aspterric acid.
