Description:
Independent transgenic line #2 containing the genetically encoded fluorescent biosensor Kinase Localization Reporter KLR-AA-AP2C1. KLR-AA-AP2C1 consists of a 35 aa section of the Arabidopsis protein PP2C-type phosphotase AP2C1, which serves as a kinase docking domain, a bipartite modified nuclear localization signal (bNLS) with two phosphosites, a nuclear localization signal (NLS), the mNeonGreen fluorophore, a T2A "self-cleaving" Thosea asigna virus 2A peptide, the mRuby3 fluorophore, and an NLS derived from simian virus 40 (SV40). The two phosphosites within the bNLS have been mutagenized to non-phosphorylable alanines. Primary transformants were identified with an epifluorescence microscope and seed was collected from plants exhibiting strong fluorescence. Transgene genotype unknown.

pKLR-AA-AP2C1 | T-DNA construct expressing a fluorescent reporter for measuring kinase activity in plant cells. This version of the reporter is a control construct with a non-phosphorylable version of the phosphoacceptor domain and a kinase docking domain derived from a region of Arabidopsis AP2C1. | CaMV 35S | mNeonGreen/mRuby3 | n/a |
