Description:
The phosphatase domain of AtPAP2 (At1g13900) is solely overexpressed on the outer membrane of chloroplasts. The coding sequence (CDS) of the C-terminal transmembrane motif of AtToc33 (At1g02280) was used to replace the C-terminal transmembrane motif of AtPAP2 (At1g13900) via overlapping PCR to create the construct P2TOC. The construct was then cloned into pCXSN vector downstream of the cauliflower mosaic virus(CaMV) 35S promoter (pCXSN-CaMV35: AtPAP2-TOC).The vector was introduced into Agrobacterium tumefaciens strain GV3101 and then transformed by the floral dip method into Col-0. Homozygous CaMV35S: AtPAP2-TOC (P2TOC) lines were selected on MS plates containing 30 g/L hygromycin
