Description:
Transgenic line expressing integrase-based, history-dependent recorder of gene expression, PhiC31 then Bxb1 history dependent target with p35S, plus PhiC31 expression under control of the SPEECHLESS promoter and Bxb1 under control of the MUTE promoter, with both integrases in the same construct. T2 seeds which are not segregated for the pSPCH::PhiC31;pMUTE::Bxb1 construct, around half of seedlings should show a PhiC31 switch from BFP to RFP in leaf epidermal cells in the stomatal lineage (descending from stomatal meristemoid) and a subsequent Bxb1 switch from RFP to GFP in only the guard cells of the stomata. All other cells should remain expressing BFP.
