| pAGO1:3xHA-AGO1-DAH | The coding sequence of AT1G48410 (AGO1) was amplified from Col-0 with an in frame N-terminal triple HA epitope sequence added. The 5' regulatory sequence of AGO1 was PCR amplified from Col-0 and ultimately ligated upstream of the HA-AGO1 coding sequence. The AGO1 3' regulatory sequence was PCR amplified from Col-0 and ultimately ligated downstream of the 5'-AGO1:HA-AGO1 sequence. Mutagenesis of the catalytic core from the wildtype DDH to DAH was performed via PCR on the Col-0 coding sequence using mutagenic oligos. | AGO1 | Hygromycin |
