| pAGO10:3xHA-AGO10-DAH | The coding sequence of AT5G43810 (AGO10) was amplified from Col-0 with an in frame N-terminal triple HA epitope sequence added. The 5' regulatory sequence of AGO10 was PCR amplified from Col-0 and ultimately ligated upstream of the HA-AGO1 coding sequence. The AGO10 3' regulatory sequence was PCR amplified from Col-0 and ultimately ligated downstream of the 5'-AGO10:HA-AGO10 sequence. The final cassette was cloned into the Gateway compatible binary vector pMDC99. Mutagenesis of the catalytic core from the wildtype DDH to DAH was performed via PCR on the Col-0 coding sequence using mutagenic oligos. | AGO10 | Hygromycin |
