The 5' regulatory sequence of AGO10 was PCR amplified from Col-0 and ultimately ligated upstream of the HA-AGO1 coding sequence. The AGO10 3' regulatory sequence was PCR amplified from Col-0 and ultimately ligated downstream of the 5'-AGO10:HA-AGO10 sequence.
