Ac-Rlc-63 line; the Ac transposase is supplied by an Ac transposon truncated at the NaeI upstream of the transposase ORF and expressed from a 35S promoter; contains a bacterial neomycin phosphotransferase (NPTII) gene expressed from a mannopine synthase (mas) promoter and followed by a mas 3' transcriptional terminator that serves as the plant transformation marker conferring kanamycin-resistance; the T-DNA vector also carries the agrobacterial tms2 gene as a negative selectable marker (that confers sensitivity to naphthalene acetamide NAM) and the Rlc gene as a visual marker (Rlc gene is a maize transcriptional activator in the anthocyanin biosynthetic pathway).

Plants containing Ac transposase can be identified by the expression of the Rlc gene, showing three distinct phenotypes detected from the time of true leaf emergence: 1) increased number of trichomes, which gives plants a hirsute appearance, 2) red to purple color due to increased anthocyanin pigment production and 3) plants grow more slowly than wild type plants.
